Prior to data collection, the assigned SPOC will message in the request thread and verify with user their preferred collection parameters. The following list are possible requests (typical settings are mentioned).
- Microscope: The one to be scheduled
- Magnification / Pixel Size: link to corresponding refined pixel sizes
- Total Exposure Dose (in e/A2): (50e/A2 default)
- Slit: (10eV default)
- Grid type
Morning of the collection, SPOC will prepare the microscope as follows:
- Set data acquisition beam to requested mag/pixel size
- Tune the energy filter at the requested collection mag
- Gain collected or not and at what dose rate
- Adjust beam with optimal dose-rate on the detector
- Direct alignments
- Objective astigmatism
- Coma correction
- Image shift calibration
- Any specific exposure related settings in the data acquisition preset
- Label autoloader & run inventory
At this point the independent user will take over the data collection. User will be emailed (via scheduling ticket) the TeamViewer login information, load list, and an overview of what settings have been adjusted on the microscope.
The independent data collector will assume control and set the file, save, run the Atlases, set up the SerialEM/EPU session, and start the automated collection.
If any problems are encountered during setup, the SPOC should be contacted via the scheduling ticket for them to troubleshoot.
